清温消热饮对小鼠LPS致炎模型血清中炎性因子的影响

摘要:
目的:了解促炎、抗炎细胞因子在小鼠LPS致炎模型中的动态变化,探讨中药方“清温消热饮”的抗炎治疗作用。方法:采用酶联免疫法测定112例用LPS致小鼠急性炎症模型治疗过程中血清中促炎因子(TNF-α、IL-1β、PGE2)、抗炎因子(IL-10)含量。结果:与LPS致小鼠急性炎症模型组比较:清温消热饮+炎症模型组在18h、24h血清中IL-1β含量明显降低,差异显著(p<0.05);清温消热饮+炎症模型组在6h、12h、18h血清中TNF-α含量明显降低,6h、18h差异显著(p<0.05),12h差异极显著(p<0.01);清温消热饮+炎症模型组血清中的PGE2含量均低于模型组,在6h、18h、24h、36h时血清中的PGE2含量明显降低,差异极显著(p<0.01);清温消热饮+炎症模型组在18h、48h血清中IL-10含量明显升高,差异极显著(p<0.01)。同时,清温消热饮+炎症模型组与地塞米松+炎症模型组在各时间段血清中TNF-α、IL-1β、IL-10、 PGE2含量无显著差异(p>0.05)。结论:清温消热饮在急性炎症模型中能有效降低血清促炎因子TNF-α、IL-1β、PGE2含量水平,提升抗炎因子(IL-10)含量水平以表现出明显的抗炎作用。

Effect of a traditional Chinease drug prescription“Qing Wen Xiao Re Yin” on Serum Inflammatory Factors in LPS-induced inflammation Model

Abstract:
objective: The study was to investigate the dynamic changes of proinflammatory and anti-inflammatory cytokines in LPS-induced inflammation model in mice and explore the anti-inflammatory effect of traditional Chinease drug prescription“Qing Wen Xiao Re Yin”. Methods: ELISA was used to detect the concentrations of serum proinflammatory cytokines (TNF-α, IL-1β, PGE2) and anti-inflammatory cytokines (IL-10) in 112 acute inflammatory models of mice induced by LPS according to the manufacturer’s protocol. Results: Compared with LPS-induced acute inflammation model group, the content of IL-1β in serum of “Qing Wen Xiao Re Yin” Inflammation model group was significantly decreased (P <0.05) at 18h and 24h.The content of TNF-αof “Qing Wen Xiao Re Yin” Inflammation model group was much lower than that in the Inflammation model group at 6h、12h、18h(P <0.05 or P<0.01) . The contents of PGE2 in the serum of “Qing Wen Xiao Re Yin” inflammation model group were significantly lower than that in the Inflammation model group at 6h, 12h and 18h (P <0.01), and the content of serum IL-10 in the “Qing Wen Xiao Re Yin” Inflammation model group was significantly lower than that in model group at 6h, 18h, 24h and 36h (P <0.01), the difference was significant (P <0.01). The levels of TNF-α, IL-1β, IL-10 and PGE2 in the serum of “Qing Wen Xiao Re Yin” Inflammation model group and dexamethasone inflammation model group had no significant difference (p>0.05). Conclusion: Qing Wen Xiao Re Yin can effectively decrease the levels of serum proinflammatory cytokines TNF-α, IL-1β, PGE2 and enhance the level of anti-inflammatory factor IL-10 in the acute inflammation model ,which showing obvious anti-inflammatory effect.
