为提高北京市兽医实验室核酸检测能力,加强实验室质量控制水平,验证PCR实验室去污染与气溶胶消除效果,本研究通过利用Z值与尧敦图统计方法对2024年北京市15家非洲猪瘟授权实验室核酸检测能力验证结果进行统计分析,对5种DNA去除剂、5种实验室常用消毒剂进行了消除核酸效果验证,使用汽化消毒机对空气中核酸气溶胶污染消除效果进行了模拟试验,验证了紫外照射对消除核酸污染的效果。结果表明:15家实验室检测结果满意。1号实验室非洲猪瘟病毒能力验证结果应给出警戒信号,5号DNA去除剂降解核酸效果明显,且通过喷洒方式可消除空气中的核酸气溶胶污染。使用35%过氧化氢汽化消毒可清除空气中核酸气溶胶污染。紫外照射过夜对消除空气中核酸气溶胶有效。 |

To enhance the nucleic acid testing capacity of veterinary laboratories in Beijing and strengthen the level of laboratory quality control, this study conducted a comparative analysis of the verification results of nucleic acid testing capabilities in 15 African swine fever authorized laboratories in Beijing for the year 2024, using Z-values and Youden plots statistical methods. The study verified the effectiveness of 5 types of DNA removers and 5 commonly used laboratory disinfectants in eliminating nucleic acids, simulated experiments on the elimination of airborne nucleic acid aerosol contamination using vaporized disinfection machines, and validated the effect of ultraviolet irradiation on eliminating nucleic acid contamination. The results indicated that the testing results of the 15 laboratories were satisfactory. Laboratory No. 1 should issue an alert signal for its African swine fever virus capability verification results, DNA remover No. 5 significantly degraded nucleic acids, and the aerosol contamination in the air could be eliminated through spraying. Vaporizing disinfection with 35% hydrogen peroxide could clear airborne nucleic acid aerosol contamination. Overnight ultraviolet irradiation was effective in eliminating airborne nucleic acid aerosols. |
