HPLC 法测定金花平喘散中盐酸麻黄碱、盐酸伪麻黄碱及苦杏仁苷的含量

摘要:
为了完善金花平喘散的质量控制方法,用高效液相色谱法对金花平喘散中麻黄药材的有效成分盐酸麻黄碱、盐酸伪麻黄碱及苦杏仁中的有效成分苦杏仁苷的含量测定进行研究。用Waters X Bridge C18(4.6mm×250mm,5μm)为色谱柱,乙腈-0.2%磷酸溶液(梯度洗脱),流速1.0mL/min,检测波长为210nm,柱温30℃,进样量10μL。结果为盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷进样浓度分别在1.114~111.4μg/mL(r=0.99984)、1.111~111.089μg/mL(r=0.99981)、3.083~154.174μg/mL(r=0.99998)范围内,峰面积与含量呈良好线性关系。样品的平均回收率(n=6)分别为94.14%、98.42%、97.70%,RSD分别为1.93%、1.12%、0.66%。该方法简单准确度高,可用于金花平喘散中有效组分的含量测定,为控制其产品质量提供依据。

Determination of Ephedrine Hydrochloride, Pseudoephedrine Hydrochloride, and Amygdalin in Jinhua Pingchuan Powder by HPLC

Abstract:
In order to improve the quality control method of Jinhua Pianchuan Powder, the content of effective components, Ephedrine hydrochloride, Pseudoephedrine hydrochloride and Amygdalin in the herb of Ephedra used in Jinhua Pianchuan Powder were determined by high-performance liquid chromatography. Waters X Bridge C18 (4.6mm × 250mm, 5μm) was used as the column, with a gradient elution of acetonitrile-0.2% phosphoric acid solution at a flow rate of 1.0mL/min, a detection wavelength of 210nm, and a column temperature of 30°C. The injection volume was 10μL. The results showed that the injection concentrations of Ephedrine hydrochloride, Pseudoephedrine hydrochloride and Amygdalin were in the range of 1.114-111.4 μg/mL (r=0.99984), 1.111-111.089 μg/mL (r=0.99981) and3.083-154.174 μg/mL (r=0.99998). There is a good linear relationship between peak area and content. The average recovery rates (n=6) were 94.14%, 98.42%,and97.70%, with RSD of1.93%, 1.12%, and0.66%. The method is simple and accurate, and can be used for the determination of effective components in Jinhua Pingchuan powder, which provides a basis for the control of product quality.
