本实验建立了一种测定大承气散中大黄酚、大黄酸、大黄素、芦荟大黄素、大黄素甲醚含量的高效液相色谱法(HPLC)。采用Hypersil GOLD C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈为流动相A、以0.1%磷酸溶液为流动相B进行梯度洗脱,检测波长为225 nm。结果显示,大黄酚、大黄酸、大黄素、芦荟大黄素、大黄素甲醚5种成分线性关系良好(r>0.999),平均加样回收率为1015%、98.1%、99.4%、99.8%和98.4%,精密度、稳定性和耐用性试验的变异系数均小于2.5%。该方法有较好的分离度、准确度,可用于测定大承气散中大黄酚、大黄酸、大黄素、芦荟大黄素、大黄素甲醚的含量,为大承气散的质量评价提供数据支持。 |

Determination of Chrysophanol,Rhein,Emodin,Aloe-emodin and Physcion in Dachengqisan by High Performance Liquid Chromatography |

Abstract: |
This experiment established a high-performance liquid chromatography (HPLC) technique to determine the content of chrysophanol,rhein,emodin,aloe-emodin and physcion in dachengqisan.The sample was separated on a Hypersil GOLD C18(4.6 mm×250 mm,5 μm)with acetonitrile-0.1% phosphoric acid in gradient elution.The detection wavelength was 225 nm. The results showed that there were good linear relationships for chrysophanol,rhein,emodin,aloe-emodin and physcion(respectively was r>0.999),the average recovery rate were 101.5%,98.1%,99.4%,99.8% and 98.4%.The RSD of precision,stability and durability was less than 2.5%.This method has good separation and accuracy, and can be used to determine the content of chrysophanol,rhein,emodin,aloe-emodin and physcion in dachengqisan, and provide data support for the quality evaluation of dachengqisan. |
