为了优化藿乌粉的提取工艺,研究以乙醇浓度、固液比、提取时间、提取次数为影响因素,2,3,5,4"-四羟基二苯乙烯-2-O-β-D-葡萄糖苷、淫羊藿苷、朝藿定A、朝藿定B含量以及干膏得率为评价指标,在单因素试验的基础上,进行L9(34)正交试验。采别采用层次分析法(AHP)、基于指标相关性的权重确定方法(CRITIC)、AHP-CRITIC综合加权法计算各评价指标的权重系数,选出最佳的多指标权重方法。利用确定出的指标权重对正交试验的结果进行综合评分,优选出藿乌粉的最佳提取工艺,并对优选出的工艺进行验证试验。结果表明AHP-CRITIC混合加权法确定出的权重系数更为科学、合理,最佳提取工艺为12倍60%乙醇提取2次,每次1.5 h。3次重复验证试验指标成分2,3,5,4"-四羟基二苯乙烯-2-O-β-D-葡萄糖苷、淫羊藿苷、朝藿定A、朝藿定B的平均质量分数分别为36.46、19.38、6.98、11.11 mg.g-1,平均干膏得率为16.74%。AHP-CRITIC法结合正交试验优选出的提取工艺经验证稳定可行,重复性好,可用于藿乌粉的制备。 |

Study on Extraction Technology of Huowu powder |

Abstract: |
In order to optimize the extraction process of Huowu powder .In this study, ethanol concentration, solid-liquid ratio, extraction time and extraction times were the influencing factors. Using the contents of 2,3,5,4"-tetrahydrostilbene-2-O-β-D-glucoside, Icariin, Epimedin A, Epimedin B and ratio of dry extraction as comprehensive evaluation indexes. L9(34) orthogonal test was carried out on the basis of single factor test. Analytic Hierarchy Process (AHP), Criteria Importance Though Intercrieria Correlation (CRITIC) and AHP-CRITIC hybrid weighting method are used to calculate the weight coefficient of each evaluation index, and the best multi index weight method is selected. Using the determined index weight to comprehensively score the results of orthogonal test. The best extraction process of Huowu powder was optimized, and the optimized process was verified.The results show that the weight coefficient determined by AHP-CRITIC hybrid weighting method is more scientific and reasonable. According to the weighting coefficient determined by comprehensive evaluation, the optimal process parameters were as follow: 12 times the amount of 60% ethanol extraction twice, each 1.5 h.The average mass fractions of the index components 2,3,5,4"-tetrahydroxystilbene-2-O-β-D-glucoside, Icariin, Epimedin A, Epimedin B were 19.38, 6.98, 11.11,36.46 mg.g-1. And the average ratio of dry extraction was 16.74% in three replicate validation tests. AHP-CRITIC method combined with orthogonal test to optimized process of extraction had been verified to be stable, feasible and reproducible, which could be used for the preparation of Huowu powder. |
