首次将维生素C碳点应用于IgY的提取工艺中,探讨不同浓度的维生素C碳点对抗草鱼IL-1β IgY二级结构及提取效果的影响。结果表明,添加终浓度6.25 mg.L-1和12.50 mg.L-1的维生素C碳点,使得IgY二级结构中α-螺旋相对含量分别较对照组提高了12.6%和5.8%,结构更稳定。添加维生素C碳点有助于卵黄中水溶性蛋白的提取并提高抗体效价,6.25 mg.L-1组的提取效果最好。此外,添加维生素C碳点能有效抑制IgY提取液中细菌的生长,实现室温条件下提取IgY。研究表明,在传统酸化水提取IgY的基础上添加维生素C碳点,可实现室温条件下规模化提取IgY。 |

Effects of vitamin C carbon points on the secondary structure and extraction effect of IgY |

In this study, vitamin C carbon point (CDs) was applied to the extraction of IgY for the first time, and the effects of CDs with different concentrations (0.00, 6.25, 12.50 and 25.00 mg.L-1) on the secondary structure and extraction effect of IgY were investigated. The results showed that: (1) Compared with the control group, the relative content of α-helix in the IgY secondary structure increased 12.6% and 5.8% respectively, with the final concentration of 6.25 and 12.50 mg.L-1 of CDs, which means its structure is more stable. (2) The addition of CDs was helpful to extract the water-soluble protein from yolk and conducive to increase the titer of antibody, in particular, the 6.25 mg.L-1 group had the best extraction effect. (3) The addition of CDs can effectively inhibit the growth of bacteria in the IgY extraction, and realize the extract of IgY at room temperature. Conclusion: Adding CDs on the basis of traditional acidification water extraction IgY can realize large-scale extraction of IgY at room temperature. |
