阿维菌素类药物残留检测衍生化研究

摘要:
本文优化了猪、牛组织及奶和奶粉中阿维菌素类药物残留检测的衍生化方法。经过对反应温度、时间及光照的系统摸索,采用N-甲基咪唑-乙腈(1+1,v/v)与三氟乙酸酐-乙腈(1+2,v/v)为衍生化试剂,经65℃避光反应15 min进行衍生化。该衍生化方法用时短、效率高,进一步提高了检测方法的灵敏度,使得乙酰氨基阿维菌素、阿维菌素、伊维菌素和多拉菌素在牛、羊奶中的定量限为0.5 μg/kg;在猪、牛组织和奶粉中的定量限为5 μg/kg。

Abstract:
An derivatization method for the determination of avermectin residues in swine and beef tissues、milk and milk powder has been developed. Based on the systematic investigation of the impact of various temperature, duration time and light, the optimized method was established using N-methylimidazole + acetonitrile(1+1,v/v) and trifiuoroacetic anhydride + acetonitrile(1+2,v/v)as the derivatization reagents and taking place at 65℃ in dark for 15 mins. This derivatization method not only get the efficient products at the short duration, but also improve the sensitivity of the method: the limit of quantification(LOQ) of eprinomectin, abamectin, ivermectin and doramectin was 0.5 μg/kg in milk and goat milk and 5 μg/kg in milk powder、 swine and beef tissues.
