三黄连散的亚慢性毒性试验研究

摘要:
探讨大鼠长期口服三黄连散的亚慢性毒性作用。将80只大鼠随机分为空白对照组、三黄连散低(2.5 g/kg·BW)、中(5.0 g/kg·BW)和高剂量组(10.0 g/kg·BW),每组20只,雌雄各半,采用混饲给药的方式连续给药30 d,每日观察临床表现,记录体重和饲料消耗量,停药后继续观察7 d。分别于给药后30d和停药后7d进行血液常规指标、血液生化指标和脏器解剖学、病理学检查。结果显示,给药期间和停药后,各试验组大鼠未出现中毒和死亡情况,临床症状、解剖学检查等均无异常变化,脏器指数、组织病理学检查结果均无异常;血液常规指标、血液生化指标结果表明该药物无毒副作用,仅见高剂量组大鼠的体重和饲料消耗量显著低于空白对照组(P<0.05)。结果表明,三黄连散给大鼠连续饲喂30天未见明显毒性反应。

Experimental Study on Subchronic Toxicity of Sanhuanglian powder

Abstract:
To investigate the subchronic toxicity of Sanhuanglian powder by oral administration in rats, eighty SD rats were randomly divided into four groups: the blank control group; the Sanhuanglian powder low-, middle-, high-dose groups [2.5, 5.0 and 10.0 g/kg·BW],20 animals (half male and female) each group. Sanhuanglian powder was given the rat by feed for 30 days, clinical observations were made daily, and body weights and food consumption were determined daily, the next observation for 7 days was conducted post treatment.Blood samples were collected on Day 30 and Day 37 respectively for measurement of blood routine and blood biochemistry indexes, gross anatomic and histological examination were conducted.The results showed that no significantly toxicological change in mortality, clinical signs, necropsy findings, organ index, and histopathological findings were found. There is no significant change in blood routine and biochemical indexes. The body weight and food consumption of rats at high-dose group were significantly lower when compared with blank control (P<0.05). In conclusion, no toxicological effect has been observed in rats treated with Sanhuanglian powde for 30 days.
