为了优化酶法辅助超声提取功劳木小檗碱的工艺,在单因素实验基础上,根据Box-Behnken中心组合实验设计原理,采用4因素3水平的响应面分析法,以小檗碱收率为响应值进行回归分析。实验结果表明,酶解最佳工艺条件为:酶解pH值3.5、酶用量10.6 mg/g、酶解温度51℃、酶解时间97 min。在此条件下,小檗碱收率为22.46 mg/g。优选的提取工艺稳定可行、收率高,为功劳木的工业化生产提供参考。 |

Optimization of Enzyme-assisted Ultrasonic Extraction Process for Berberine from Mahonia bealei |

Abstract: |
To optimize enzyme-assisted ultrasonic extraction process of berberine from Mahonia bealei. Experimental factors and their levels were determined by one-factor tests. Subsequently, the Box-Behnken experimental design with 4 factors and 3 levels was performd, and the factors influencing the extraction process were estimated by means of regression analysis with the berberine yield as the response value. The optimal conditions were as follows: pH 3.5, cellulase dosage 10.6 mg/g, enzymolysis temperature 51℃, enzymolysis time 97 min. Under this condition, the berberine yield was up to 22.46 mg/g. The optimized extraction technique is stable and feasible, with a high extraction rate, providing reference for industrialized production of Mahonia bealei. |
