HPLC法测定枇杷叶中齐墩果酸和熊果酸含量

摘要:
目的:建立枇杷叶中齐墩果酸和熊果酸含量的高效液相色谱(HPLC)测定方法。方法:选用色谱柱Agilent Ecilpse plus C18柱(250mm×4.6mm,5μm),流动相:乙腈-0.1%氨水(70:30)磷酸调pH=8.0,流速为0.8ml/min,检测波长210nm,柱温25℃。结果:显示齐墩果酸和熊果酸能达到基线分离,分离度R>2.5,齐墩果酸在0.19854~7.9412μg、熊果酸在0.21218~8.48704μg范围内呈良好的线性关系。结论:试验证明该方法操作简单,灵敏度高,重现性好,可用于枇杷叶药材的质量控制。

Abstract:
Objective: To establish an HPLC method for the determination of Oleanolic Acid and Ursolic Acid in loquat leaf. Methods: HPLC was carried out on Agilent Ecilpse plus C18 column(250mm×4.6mm,5μm) at 30℃ using acetonitile-0.1%ammonium water (70:30, Phosphoric acid pH to 8.0) as mobile phase with a flow rate of 0.8 ml/min and the UV detection wavelength was set at 210 nm.Result: Oleanolic Acid and Ursolic Acid can reach the baseline separation, degreeSofSseparation(R) was more than 2.5. Oleanolic acid was linear in the range of 0.19854~7.9412μg. Ursolic acid was linear in the range of 0.21218~8.48704μg. Conclusion: The method is simple, convenient, accurate, and can be used for the quality control of loquat leaf.
