建立同时测定复方蒲公英颗粒中秦皮乙素和咖啡酸两种成分的HPLC检测方法。采用RP-HPLC法,色谱柱为WondaCract ODS-2(250mm×4.6mm,5μm);流动相为甲醇-0.6%冰醋酸水溶液(20:80),体积流量1.0mL/min;检测波长326nm,柱温为25℃。两种成分达到基线分离,在进样浓度分别为0.02mg/mL-0.32mg/mL 和 0.004mg/mL-0.064mg/mL范围内均有较宽的线性范围和良好的线性关系(R2=0.9999),回收率分别为100.08%和99.69%。本方法准确、灵敏、可靠,重现性较好,可用于复方蒲公英颗粒的质量控制。 |

Abstract: |
Objective To establish an HPLC method for assay of Esculetin and Caffeic acid in the compound dandelion particles. Method: RP-HPL.A WondaCract ODS-2 column(250μm×4.6μm,5μm)was used. The mobile phase was methaonol – 0.6% glacial acetic acid(20:80) with volume flowrate 1.0mL/min; gradient elution and the detection, the wavelength was 326nm, the column temperature was 25℃. Result: Two component reach baseline resolution, and each component has good linear range and linear relation within the ranges of 0.02mg/mL-0.32mg/mL and 0.004mg/mL-0.064mg/mL (R2=0.9999). The recovery rate of Esculetin and Caffeic acid were 100.0% and 99.69%,respectively. Conclusion: The method is practicable in assay of Esculetin and Caffeic acid in the compound dandelion granules |
