为比较二乙烯亚胺(BEI)和甲醛对猫杯状病毒(Feline calicivirus,FCV)的灭活条件,本试验采用终浓度分别为0.05%、0.1%、0.2%和0.3%的甲醛和终浓度分别为0.5%、1%、2%和3%的BEI在37℃对FCV灭活6,12,24,36,48 h,通过微量滴定法检测灭活病毒滴度变化,于F81细胞盲传2代验证病毒灭活效果,采用巢式PCR和ELISA法检测病毒抗原完整性。结果显示:37℃下作用48 h,1%的BEI可灭活FCV且灭活效果好于0.1%~0.2%甲醛。本研究结果可为FCV灭活疫苗的灭活工艺研究提供参考。 |

Comparision of Inactivation Effect of Binary Ethylenimine and Formaldehyde on Feline calicivirus |

Abstract: |
To study the best inactivated condition of Feline calicivirus (FCV)by binary ethylenimine (BEI) and formaldehyde, a strain of FCV were inactivated by BEI withSfinalSconcentrationsSofS0.5%, 1%, 2%, 3% and formaldehyde with 0.05%, 0.1%, 0.2%, 0.3%, incubated at 37℃ for 6, 12, 24, 36, 48h. The viralStiters after inactivation were detected by Smicrotitrimetry and inactivation effect were validated by two blind passages on F81 cells. Novel nested PCR and ELISASmethod were used to detect the antigen integrity. The results showed that 1% BEI presented better inactivated effect than 0.1%~0.2% formaldehyde and could inactivate feline calicivirus at 37 ℃ for 48h. |
