为了提高猪瘟活疫苗的外源性兔出血症病毒(RHDV)的检测敏感性,进行了RHDV对家兔感染途径的致死性比较试验,以便筛选出具有较高致死性的感染途径。本试验将RHDV通过皮下、静脉两种途径感染家兔,比较了家兔两种感染途径的半数致死量(LD50),同时对1批污染RHDV的脾淋源猪瘟活疫苗采用皮下、静脉两种途径注射家兔进行RHDV检测。结果表明,静脉途径对家兔的LD50是皮下途径的126倍;污染疫苗的检测应用也表明家兔感染的静脉途径比皮下途径更加敏感,提高了猪瘟活疫苗外源性RHDV污染的检出率;RT-PCR证实了注苗死亡家兔的肝脏组织含有RHDV。该试验也为《中国兽药典》的修订提供了可靠的数据依据。 |

Lethality Comparison and Preliminary Application of Two Infection Routes of Rabbit Haemorrhagic Disease Virus |

To improve the testing sensitivity of Rabbit Haemorrhagic Disease virus (RHDV) contaminating in classical swine fever vaccine, lethality comparison of two Infection routes of RHDV were carried out to choose a much more fatal route.Comparing the mean of lethal dose(LD50) of rabbits infected with RHDV through intravenous and subcutaneous route, and detecting RHDV contaminating in a batch of classical swine fever vaccine with the two routes, the results showed that LD50 of rabbits infected through intravenous route is 126 times than subcutaneous route, and infection of the intravenous route gave more satisfactory result, which improved detection rate of RHDV contaminating in the vaccine. The presence of RHDV?in liver tissue of the died rabbits intravenously injected with the contaminated vaccine was confirmed by reverse transcriptase polymerase chain reaction (RT-PCR) .The test also provided reliable data for revising China Veterinary Pharmacopoeia. |
