本实验旨在研究发病鲤鱼病原菌的特性及种类,初步了解肺炎克雷伯氏菌在鲤鱼鱼体中的发病机制和防治药物的筛选。首先对发病鲤鱼进行细菌分离纯化,并进行革兰氏染色反应。经过各种生理生化反应测定单菌落特性之后进行细菌回归实验。随后进行药敏实验,对注射菌液后发病的鱼进行细菌再分离。实验得到该纯菌为革兰氏阴性菌,并且与七叶苷及其它17种指标反应呈阳性,与戊二酰-甘氨酸-精氨酸-AMC及其它11种指标反应呈阴性。注射该菌液48h后鱼体出现相同病症。药敏试验结果中得到该菌对亚胺培南等6种药物高度敏感。再分离细菌实验中得到相同的细菌。根据《伯杰氏细菌鉴定手册》,最终确定该菌为肺炎克雷伯氏菌。本实验首次从鲤鱼体内分离出肺炎克雷伯氏菌,并得到几种高度敏感的药物,阐述了发病症状,为治疗和预防该菌对鲤鱼的危害提供可靠实验数据和依据。 |

Isolation and Identification of Klebslella pneumoniae from Carp |

Abstract: |
This experiment was to study the characteristics and types of the pathogenic bacteria in the sick carp, which could preliminarily understand the pathogenesis and the screening of drug about Klebsiella pneumoniae in carp.This experiment first separated and purified single colonies from the onset of carp and the bacteria were done the Gram stain experiment.With various physiological and biochemical reacting,the experiment determined characteristics of the single colony bacterial and then did the regression experiment. Followed by drug sensitivity test,the experiment would separate the bacteria from fish who had injected bacteria liquid. The result was that this bacteria were gram negative .The purified bacteria were reacted with Esculin and other 17 kinds of indexs, and they were positive reaction;while they were negative reaction with Glutary-Glycine-Arginine-AMC and other 11 kinds of indexs.The regression experiment proved that the fish injected the bacteria liquid and appeared the same disease.drugs.Then physicochemical properties of the bacteria isolated from bacteria experiment were the same.According to Berger's Bacterial Identification Manual,the bacteria were Klebsiella pneumoniae.Summarize the above results, we conclude that this test determined that the bacterium was Klebsiella pneumoniae, as well as drug sensitivity identified some kinds of drugs which were highly sensitive to the bacterium.This study provides the reliable theory basis and data for the treatment and prevention of the carp’s diseases caused by bacteria.
