为寻求抗ALV-J感染雏鸡用药物,通过CCK-8试剂盒检测细胞活力以确定利巴韦林、病毒灵和黄芪多糖对DF1细胞的最大安全浓度,用ELISA试剂盒检测ALV-J P27抗原表达量,以确定上述抗病毒药物体外抗ALV-J活性。结果显示,利巴韦林、病毒灵和黄芪多糖对DF1细胞的最大安全浓度分别为0.0391、0.0625和0.0625 mg/mL。在安全范围内,利巴韦林对P27的表达量具有明显的抑制作用,黄芪多糖在0.0039~0.0625 mg/mL安全浓度范围内明显抑制P27的表达,病毒灵在安全范围内对P27的表达基本上没有抑制作用。试验表明,利巴韦林和黄芪多糖对ALV-J的增殖有明显的抑制作用,有望作为该病净化的辅助性防制药物。 |

The Activity Experiment of Ribavirin, Moroxydine and Astragalan Against ALV-J |

To acquire the anti-ALV-J drug,P27 expression quantity influence of Ribavirin, moroxydine and astragalan on ALV-J (NX0101) are researched by the CCK 8 kits cell detected for the maximum safe concentrations of the three antiviral drugs on DF1 cell and ALV-J P27 antigen ELISA test kit for P27 expression quantity.The results show that the maximum safe concentrations of Ribavirin, moroxydine and astragalan on DF1 cell are 0.039, 0.0625 and 0.0625 mg/mL. The obvious inhibitory concentration of Ribavirin for the P27 expression quantity in the the scale of safe concentrations is 0.0391 mg/mL. The concentration range of astragalan inhibiting the expression of ALV-J P27 are 0.0039~0.0625 mg/mL. However, varied concentration of moroxydine in the research has no inhabitory effect on P27 expression quantity. The aforementioned results show that ribavirin and astragalan can inhabit the propogation of ALV-J, providing a perfect prospect for the ALV-J eradication in chinken in the future. |
