乙酰甲喹及其主要代谢物在鸡体内的残留消除规律研究

摘要:
建立了高效液相色谱串联质谱(HPLC-MS/MS)法用于乙酰甲喹及其6种主要代谢物的检测。将65只健康白羽鸡分为两组,10只为空白对照组,其余为试验组。试验组施以20 mg/kg b.w.灌胃乙酰甲喹悬浊液,1 d 2次,连续3 d。在给药结束后2、4、6、12、16、24、30、36、48、72、120 h分别宰杀5只鸡,采集血液和可食性组织(肌肉、肝脏、肾脏和皮脂)样品。结果表明:乙酰甲喹原药在组织和血浆中迅速消除;乙酰甲喹的代谢物广泛存在于鸡的可食性组织和血浆中;代谢物的残留消除过程较为复杂,代谢物在肌肉和肝脏中残留量较多,在皮脂中残留时间最长。研究结果将有助于揭示乙酰甲喹在鸡体内的残留消除规律。

Residue Depletion of Mequindox and Its Main Metabolites after Gavage Administration of Mequindox in Chickens

Abstract:
Sixty-five healthy chickens were divided into two groups, ten of them in control group and the others in test group. The test one was received gavage administration twice daily at doses of 20 mg/kg b.w. for 3 days, and five of them were sacriced at 2, 4, 6, 12, 16, 24, 30, 36, 48, 72, 120 hours after treated at random, respectively. Plasma and edible tissues (muscle, liver, kidney and fat) samples were collected for each chicken. HPLC-MS/MS system was used to sample detection, WinNonlin 5.2.1 was chose to data analysis. The method built in this study was simple, precise and sensitive. The results of animal text showed that the metabolites of mequindox existed extensively in chicken edible tissues and plasma. The depletion process was very complicated that the higher residue levels were found in chicken muscle and liver while the longest residue time was discovered in chicken fat. Those results could help us to understand the residue depletion of mequindox and its major metabolites in chickens.
