山羊传染性胸膜肺炎支原体C87001株菌种纯化克隆研究

摘要:

为获得纯净山羊传染性胸膜肺炎支原体生产用菌种，将山羊传染性胸膜肺炎C87001株冻干肺组织（1980 年），通过采用绵羊肺炎支原体代谢抑制试验结合固体培养基挑取单个克隆菌落方法，进行纯化。结果表明，制苗用C87001株冻干肺组织（1980 年）纯化后克隆株1～15代菌种均纯净、每代活菌滴度均≥109.0CCU/ml，经菌落形态和PCR鉴定，获得克隆纯化的F5代山羊传染性胸膜肺炎支原体生产用菌种。该菌种经本动物回归试验4/4发病；按规程制备灭活疫苗免疫山羊攻毒保护为4/4（100%），对照组3/3发病死亡。

Abstract:

To pure caprine infectious pleuropneumonia strains of mycoplasma, the caprine infectious pleuropneumonia strains of mycoplasma inactivated C87001 plants freeze-dried lung tissue (1980). By adopting the sheep mycoplasma pneumoniae metabolic inhibition test in combination with solid medium from a single clone colony method, cloning and purification. The results showed that the seedling with C87001 plants freeze-dried lung tissue after purification cloning plants (1980) 1~15 strains are pure, each generation of living bacterium drop of 109.0 CCU/ml or higher, the colony morphology and PCR identification, for cloning and purification of F5 generation of caprine infectious pleuropneumonia strains of mycoplasma bacteria, through this animal regression test 4/4 onset; According to the rules of the preparation of inactivated vaccine immune goats attack poison protection for 4/4 (100%), control group 3/3 disease death.
