本试验利用杆状病毒双表达系统构建高致病性猪繁殖与呼吸综合征病毒(HP-PRRSV) N蛋白和GP5蛋白的重组杆状病毒Bac-N-GP5,对重组病毒进行间接免疫荧光、Western blot分析以及动物免疫试验。结果表明:重组杆状病毒中表达了重组N和GP5蛋白,重组杆状病毒免疫小鼠后产生了抗PRRSV GP5蛋白和N蛋白的特异性抗体。本试验成功构建共表达PRRSV N蛋白和GP5蛋白的重组杆状病毒,为下一步应用该重组杆状病毒作为基因工程亚单位疫苗预防PRRSV感染奠定基础。 |

Abstract: |
In order to combine the humoral immunity of GP5 protein and cellular immunity of N protein of highly pathogenic porcine reproductive and respiratory syndrome virus, Identification by indirect immunofluorescence, western blot, results showed that recombinant baculoviruses Bac-N-GP5 co-expressed the N and GP5 proteins. After immunization in BALB/c mice with cell lysates treated by aluminium adjuvant, Bac-N-GP5 induced strong N and GP5-specific ELISA antibodies. These results suggested that the recombinant baculoviruses can be a potential genetic engineering vaccine against PRRSV infections. |
