本研究使用间接免疫荧光方法(IFA)和过氧化物酶单层试验(IPMA)测定猪瘟兔化弱毒的组织半数感染量(TCID50)。优化了两种方法的一抗使用浓度;筛选了IPMA方法中敏感、易辨识的酶作用底物。优化后的IFA和IPMA方法测定猪瘟兔化弱毒疫苗半成品和成品的TCID50,可以作为兔体感染量(RID)和猪体半数保护量(PD50)效力检验标准的补充,应用到猪瘟兔化弱毒活疫苗生产中,加强质检、质控力度。 |

Abstract: |
:Indirect immunofluorescent assay (IFA)and Immunoperoxidase monolayer assay(IPMA) were used to titrate the Tissue Culture Infectious Dose 50(TCID50) of Hog cholera virus lapinized Chinese strain(HCLV). Optimized antibody dilution and Peroxidase Substrate. IFA and IPMA can titer TCID50 of finished and semi-finished products of HCLV vaccines.
