猪伪狂犬病活疫苗(Bathar-K61株)免疫佐剂的筛选

摘要:为筛选到一种适合猪伪狂犬病活疫苗的免疫佐剂,本研究通过稳定性试验、病毒效价测定、安全性试验、效力试验对5种不同类型免疫增强佐剂(AD01、AD02、AD03、AD04和AD05)进行了比较筛选。稳定性试验结果:AD01、AD03、AD04和AD05等4种佐剂稀释疫苗后6hr不分层,表现出良好的稳定性,而AD02佐剂稀释疫苗1个小时后出现明显分层;病毒效价测定结果表明,AD01、AD03、AD04等3种佐剂稀释后疫苗滴度为106.25~6.50TCID50/100uL,与对照组病毒滴度无差异,对病毒效价没有干扰,而AD05佐剂稀释疫苗后,测得的病毒滴度为103.5~5.17TCID50/100uL,低于对照组病毒滴度,对病毒效价有干扰;安全性试验表明,用AD01佐剂稀释疫苗免疫猪群后,猪只体温明显升高,平均体温为40.3?0.2℃,有20%(2/10)的猪只出现呕吐,40%(4/10)的猪只食欲下降和精神不振的临床症状,而AD03、AD04佐剂稀释疫苗免疫猪群后,临床表现无异常,与对照组一致;效力试验结果表明,剩余的AD03、AD04两种佐剂稀释疫苗后在第7天均可检测到中和抗体,第60天抗体的中和效价平均值达到高峰,分别为69.8?3.3和69.5?2.8,高于对照组的57.8?7.0。佐剂组产生抗体的中和效价、整齐度、持续时间均优于普通稀释液稀释疫苗的猪群。对比结果说明,AD03、AD04两种佐剂均适用于猪伪狂犬病活疫苗(Bartha-K61),为提高猪伪狂犬病活疫苗(Bathar-K61株)的实际使用效果进行了有益的探索。

Abstract:
Abstract: To screen an appropriate adjuvant for swine pseudorabies live vaccine, 5 different types of adjuvant (AD01, AD02, AD03, AD04 and AD05) were compared and evaluated through stability test, testing of virus titer, safety test and potency test.The result of stability test shows that, the vaccine diluted by AD01、AD03、AD04 and AD05 was not layered after 6hr, demonstrating good stability, the vaccine diluted by AD02 was obviously layered after 1 hour;The testing result of virus titer shows that, the titer of vaccine diluted by AD01、AD03、AD04 is 106.25~6.50TCID50/100uL,which was not different with the titer of control group, demonstrating no interference to the virus, the titer of vaccine diluted by AD05 was 103.5~5.17TCID50/100uL, which was lower than the control group, demonstrating interference to the titer of virus; The safety test shows, after vaccinated with vaccines diluted by AD01, the temperature of the pigs increased significantly, the mean value is 40.3?0.2℃,20%(2/10) pigs vomited, 40%(4/10) pigs showed clinical symptoms including loss of appetite and depression. The pigs vaccinated with vaccines diluted by AD03、AD04 did not show any exceptional clinical signs, which is same as control group. The potency test shows, the neutralizing antibody can be checked at the 7th day after vaccinated with the vaccine diluted with AD03, AD04, the neutralizing titer achieved the peak at the 60th day, the mean values were 69.8?3.3 and 69.5?2.8 respectively, which were higher than the control group (57.8?7.0). Neutralizing titer, uniformity and duration of the antibody occurred in the adjuvant groups were better than the control group diluted with normal dilution. The comparison test shows that both AD03 and AD04 can be used as adjuvant of swine pseudorabies live vaccine(Bathar-K61), which provides a profitible exploration for the improvement of swine pseudorabies live vaccine(Bathar-K61 strain) in field.
