两种磺胺氯吡嗪钠溶液剂灌服给药在鸡体中的比较药动学研究

摘要:
24只健康肉鸡随机分成2组,每组12只(公母各半),采用单周期平行设计,分别进行单剂量(60 mg/kg bw,以磺胺氯吡嗪钠计)经口灌服复方和单方磺胺氯吡嗪钠溶液的药动学研究。给药后按预定时间采集血样,采用HPLC法测定血浆中药物含量。实测血药浓度-时间数据,采用Kinetica药动学分析软件计算药动学参数。结果显示,鸡单剂量灌服磺胺氯吡嗪钠溶液后,主要药动学参数如下:T1/2β为9.39 h,MRT为26.57 h,Tmax和Cmax分别为7.33 h和64.69 μg/mL,ClB为0.52 mL/(min?kg),Vz为0.43 L/kg,AUC为1954.28 μg/mL?h。鸡单剂量灌服复方磺胺氯吡嗪钠溶液后,主要药动学参数如下:T1/2β为8.24 h,MRT为26.37 h;Tmax和Cmax15.50 h和71.43 μg/mL;ClB为0.51 mL/(min?kg),Vz为0.36 L/kg,AUC为2125.81 μg/mL?h,相对生物利用度为108.78%。结果表明,与磺胺氯吡嗪钠溶液相比,复方磺胺氯吡嗪钠溶液灌服后,除达峰时间和峰浓度明显延长或增高(P <0.01)外,其他药动学参数均无显著差异(P>0.05)。

Abstract:
24 healthy broiler chickens were randomly divided into two equal groups, A single dose of parallel test design was used. The pharmacokinetics of compound sulfaclozine sodium(SPZ) solution and sulfaclozine sodium solution were studied in broiler chickens by oral route (60 mg/kg b.w.). The plasma concentrations of the SPZ were determined by high-performance liquid chromatographic methods with UV detection at a wavelength of 274 nm, and pharmacokinetic parameters were calculated by kinetica program. The main pharmacokinetic parameters of SPZ solution were as follows: T1/2β max 7.33 h, Cmax 64.69 μg/mL, ClB 0.52 mL/(min?kg) and Vz 0.43 L/kg; The main pharmacokinetic parameters of compound SPZ solution were as follows: T1/2β 8.24 h, MRT 26.37 h, Tmax15.50 h, Cmax71.43 μg/mL, ClB 0.51 mL/(min?kg), Vz 0.36 L/kg, and the relative bioavailability was 108.78%. Statistical analyses revealed that, compared with sulfaclozine sodium solution, significant delay in Tmax and increase in Cmax were observed in the group, which was administered compound SPZ solution by oral route, other parameters of pharmacokinetics were not significant differences.
