采用热加速稳定性试验,以ELISA抗体效价为考察指标,确定猪瘟病毒阳性血清的贮存有效期。拟合抗体效价随时间失活的线性回归方程及不同温度下失活速率常数k值的线性回归方程。从较高温度下的k值估算较低温度下的k值。由k值计算该血清在-20℃下的贮存有效期。结果表明,该血清在-20℃下的有效期约为16.8年。1年的持续稳定性监测结果也与热加速稳定性试验预期一致。该阳性血清可以用于制备猪瘟病毒阴、阳性血清国家参照品。 |

Accelerated Thermostability Test of the Antiserum for Classical Swine Fever Virus |

Abstract: |
Using accelerated thermostablility test to evaluate the valid storage period of the antiserum for classical swine fever virus with kinetic analysis of ELISA antibody titers. The linear regression equations fitting the antibody titers over time and fitting the k parameters over temperatures were conducted. K parameter of lower temperature was estimated from k parameter of higher temperatures. So, the validity of the serum stored at -20 ℃ was calculated by the k parameter. The results showed that the serum stored at -20 ℃ would be valid for about 16.8 years. Results of continuous stability monitoring for one year were also in accordance with that expected by the accelerated thermostablility test. It could be concluded that the antiserum could be used for the preparation of national reference materials for negative and positive antisera of classical swine fever virus. |
