目的:建立快速、稳定的家兔血浆中地克珠利的提取方法,为进一步研究家兔组织中地克珠利的残留量提供基础。方法:采用HPLC法以妥曲珠利为内标,N,N-2甲基甲酰胺及乙腈联合沉淀法提取家兔血浆中地克珠利,色谱柱为Hypersil ODS (2.5 μm,250 mm?4.6mm),流动相为乙腈:水:三乙胺:冰乙酸=55:45:0.2:0.12(v/v),流速为1.0ml/min,紫外检测波长为240 nm。结果:在0.156~20.0 μg/mL浓度范围内,地克珠利的峰面积与其浓度呈现良好的线性关系(r2=0.9992);回收率为98.9%~102.4%;方法精密度RSD最大值为6.8%;最低检测限为0.078 μg/mL。结论:DMF及乙腈联合沉淀法适用于家兔血浆中的地克珠利的提取。 |

HPLC method for the determination of diclazuril in rabbit plasma |

Abstract: |
Objective: To establish a rapid and stable tissue extraction method for the determination of diclazuril residues in rabbit plasma. Methods: Diclazuril and Toltrazuril(internal standard)were added in blank rabbit plasma, extracted by N,N-dimethylformamide (DMF)/acetonitrile precipitation method and determined by HPLC(Hypersil ODS-C18).The mobile phase consisted of a mixture of tirethylamine/water/triethylamine/aceticacid (55:45:0.2:0.12) at the flow rate was 1.0ml?min-1;The ultraviolet detection wavelength was set at 240nm.. Results: The method was proven to be linear over a plasma diclazuril concentration range of 0.156 to 20.0 μg/mL with a mean correlation coefficient of 0.9992; The recovery of diclazuril from rabbit plasma was found to be 98.9~102.4%; The max RSD of method precision was found to be 6.8%; The detection limit was 0.078 μg/mL Conclusions: DMF/acetonitrile precipitation method is suitable for the extraction of diclazuril in rabbit plasma. |
