本研究选用pNZ8149-COE NZ3900重组乳酸菌作为绿色荧光蛋白(green fluorescent protein,GFP)的表达载体,制成活菌制剂,口服免疫小鼠,以GFP基因作为标记跟踪重组菌在小鼠体内的定位,并利用荧光定量PCR技术对各部位菌定量分析。结果表明,由本研究构建的重组乳酸乳球菌PEDV-COE-GFP NZ3900可较长时间定植于胃肠黏膜表面,为基因工程重组乳酸菌口服疫苗的研究奠定了基础。 |

Studies on colonization of recombine Lactococcus lactis with PEDV COE gene in mice |

Abstract: |
For this research, pNZ8149-COE were chosen as the expression vector of GFP(green fluorescent Protein).The BALB/c mice were orally dosed with approximately GFP-labeled Lactococcus lactis NZ3900.The lactis in the body of the mice was detected by FQ-PCR. The results show that the recombined lactic acid bacteria PEDV-COE NZ3900 could field planting gastrointestinal tract for longer time, which has provided a ground for research of genetic engineering recombined lactic acid bacteria oral vaccine. |
