本试验采用方瓶存放BHK21细胞悬液,在2-8℃冰箱经过不同时间段的存放,按常规传代方法进行传代。对比不同存放时间复苏时的驯化次数及细胞活力,寻求最佳复苏时间以指导生产。同时与常规从液氮中复活细胞周期及达到扩繁细胞数做对比,以说明存放悬液的优势所在。结果表明,方瓶细胞悬液最长存放60天,经4-5代驯化,细胞形态恢复,细胞数量也与做悬液前细胞数量相差无几,并能稳定传20代以上。 |

Abstract: |
Preserving Solution of BHK21 cells in square bottles for different time under 2-8℃ to culture by conventional way.We can find the better time of recovery of cells to guide production by comparing training times and vitality of the BHK21 cells.In the same time, the superiority of preserving Liquid of BHK21 cells in square bottles may indicate by comparing recovery of the cells in liquid nitrogen jars. The results showed that the shape and quantity of preserving Solution of BHK21 cells in square bottles in different time under 2-8℃ for 60 days can resume after cultured 4~5 generations, and may culture 20 generations steadly. |
