比色法测定四味健蚕增茧液中总黄酮的含量

摘要:
[摘 要] 建立四味健蚕增茧液中总黄酮含量测定的方法。采用硝酸铝、亚硝酸钠和氢氧化钠为显色剂,利用其与四味健蚕增茧液中黄酮类物质生成红色铝螯合物,以芦丁作为对照品,在510nm波长处进行比色测定,通过吸收度计算得到四味健蚕增茧液中总黄酮的含量。在8.64-12.96μg/ml范围内,吸收度与浓度呈良好的线性关系(r2=0.9997),平均回收率为98.07 %;RSD% = 2.42%。该法结果稳定,重现性好,可作为四味健蚕增茧液中总黄酮的含量测定方法。

Abstract:
Abstract: Aim: to develop a method to accurately determine the total flavonoid content in Siwei Jiancan Zengjian liquid. AlNO3-NaNO2-NaOH is chosen as the chromogenic agent for its chelating property with flavonoids, forming characteristic red flavonoid chelate aluminum. The absorbance of light of 510nm wavelength is then measured and compared with rutin as the reference standard in order to determine the total flavonoid content in the sample. The result demonstrates good linear relationship between absorbance and concentration within the range of 8.64-12.96μg/ml total flavonoid concentration, with average recovery rate of 98.07%;RSD% = 2.42%。 Conclusion: This method of analysis is consistent and reproducible, and can be used to determine the total flavonoid content in Siwei Jiancan Zengjian liquid.
