通过水质和血清对比试验,结果表明:以水质电导率低于0.5µs/cm;猪血清总蛋白不低于49g/L,胆固醇不高于1.66mmol/L为标准,制备的改良Frey培养基和支原体培养基活菌滴度均达109CCU/ml。对滑液支原体GX11-T株及猪鼻支原体BTS-7 株1~5代对数生长期培养物检测,均达109CCU/ml。改良Frey培养基与支原体培养基经反复冻融8次;或改良Frey培养基经37℃保存24小时; 支原体培养基37℃保存8小时均不影响灵敏度。改良Frey培养基在鸡胚成纤维细胞及鸡胚尿囊液中,最小检测量分别为2CCU/ml和4CCU/ml;支原体培养基在BHK21细胞中最小检测量为8CCU/ml。改良Frey培养基和支原体培养基4℃有效期为1月,-20℃为12月。 |

Abstract: |
The result of different kinds of water and serum is confirmed that the tires of medium made by the conductivity rate of water with not more than 0.5Чs/cm 、general protein of serum with not less than 49 g/L and cholesterol with not higher than 1.66mmol/L reaches 109CCU/ml。1~5 logarithm phase culture of Mycoplamas gallisepticum and Mycoplamas synoviae is attained 109CCU/ml by detection with medium。After 8 times by repeating freezing and thawing between modified Frey medium and Mycoplamas medium,or modified Frey medium in 37℃ for 24 hours and Mycoplamas medium in 37℃ for 8 hours ,the sensitivity of medium retains invariability. the limit tests for modified Frey medium is 2CCU/ml and 4CCU/ml in allantoic fluid of chick embryo and chicken embryo fibroblasts respectively.and the limit tests for Mycoplamas medium is 8CCU/ml in BHK21 cell lines.the expire date between modified Frey medium and Mycoplamas medium is 1 month in 4℃,or 12 months in -20℃. |
