连翘酯苷对体外培养细胞的毒性研究

本研究探讨了连翘酯苷对体外培养细胞的毒性,以为人们更好的研究利用连翘酯苷提供实验依据。实验采用细胞病变法观察连翘酯苷对猪肾细胞(PK-15)、猴肾细胞(marc-145)、鸡胚肾细胞(CEK)、鸡外周血淋巴细胞(chLc)等四种细胞的形态损伤;用MTT法检测连翘酯苷作用这四种细胞后的细胞活性和细胞数量的变化。据这两种方法的结果计算出连翘酯苷对这四种细胞的半数致死量。实验结果显示高剂量的连翘酯苷可致细胞发生病变,而低剂量组的细胞形态优于对照组。经SPSS16.0统计,连翘酯苷对PK-15、marc-145、CEK三种细胞的半数致死量分别为137.8 μg/mL、87.1 μg/mL和384.4 μg/mL,对chLc应用到中华人民共和国兽药典(2000年版)规定药量的10倍剂量未发现致死性。因此,适当合理的连翘酯苷用量对体外培养的细胞没有毒性,而有一定的维持细胞形态的营养作用和刺激细胞增值的作用。

Study on the Toxicity of Forsythiaside on Cell Culture

Abstract:
Forsythia is one of the most important heat-clearing and detoxicating drugs in Chinese medicine. Aiming to investigate its toxicity on cell culture, four kinds of cells were selected, namely, pig kidney cells (PK-15), monkey-kidney cells (marc-145), chicken embryo kidney cells (CEK) and chicken lymphocytes (chLc). Through this research, we hope to offer experimental basis for exploring and making use of Forsythiaside even better. Observe the cytopathic effect (CPE) of all of the cells mentioned above except chLc by inverted microscope; Examine the cytoactive and the change of cell quantities by MTT. Based on the results of these two methods, attempt to calculate the median lethal dose (LD50) of Forsythiaside. The results show: High doses of Forsythiaside might render CPE, while the low monitoring team's cell-morph was superior to the control group. According to the statistical analysis by SPSS16.0, the LD50 of the Forsythiaside on PK-15, marc-145 and CEK was 137.8 μg/mL, 87.1 μg/mL, 384.4 μg/mL. 95% confidence interval was 128.7 μg/mL -151.7 μg/mL for PK-15, 73.7 μg/mL -100.4 μg/mL for marc-145 and 334.8 μg/mL -469.2 μg/mL for CEK. There was no damage by using 10 times Forsythiaside than the People's Republic of China beastly pharmacopoeia on chLc. By these experiments we can get the conclusion: Appropriate and rational drug concentration can sustain cellular form and stimulate cell increment rather than cause toxicity on the four types of cells we have selected.
