兽用穿琥宁注射液在犬体内的药代动力学研究

摘要:

为考察兽用穿琥宁注射液经颈部背侧皮下注射在靶动物比格犬体内的药动学行为。本试验取雌雄各半的比格犬8只，按照5mg/kgBW剂量经比格犬颈部背侧皮下注射兽用穿琥宁注射液1次，分别在给药前及给药后0.083、0.25、0.50、1、2、4、6、8、12、16、24、36、48、60、96h采血2ml，离心取上层血浆，采用HPLC加内标法，检测血浆中脱水穿心莲内酯琥珀酸半酯（DAS）含量，应用药动学软件WinNonlin6.4进行相关药动学参数的计算。结果显示：比格犬血浆中DAS的Cmax为21.51μg/ml，Tmax为2.25h，T1/2为21.51h，AUC0-∞为29480.36min?μg/ml。结果表明：本试验分析方法准确、重现性好，比格犬皮下注射穿琥宁注射剂后，在体内达峰时间较快，半衰期较长，为临床制定合理的给药方案提供试验依据。

Abstract:

To explore the pharmacokinetic behavior of chaunhuning injection when administered subcutaneously to the dorsal neck of the target animal, the beagle dog.In this experiment, 8 male and female beagle dogs were given a subcutaneous injection at the back of the neck at a dose of 5 mg/kgBW. 2 ml of blood was collected before and after administration at 0.083、 0.25、0.50、1、2、4、6、8、12、16、24、36、48、60 and 96 h, respectively.The content of DAS in plasma was determined by the HPLC and internal standard method.The pharmacokinetic parameters were calculated using WinNonlin6.4.The results indicated that the Cmax of DAS in beagle plasma was 21.51μg/ml, Tmax was 2.25h, T1/2 was 21.51h, and AUC0-∞ was 29480.36min?μg/ml.The results demonstrated that the analytical method was accurate and reproducible.After subcutaneous injection of chuanhuning injection in beagle dogs, the peak time in vivo was quicker and the half-life was longer, providing an experimental basis for the clinical formulation of reasonable drug administration.
