高效液相色谱法测定饲料原料葡萄糖胺盐酸盐含量

摘要:
本实验建立高效液相色谱法测定饲料原料葡糖糖胺盐酸盐含量。采用ODSC18柱(150 mm×4.6 mm,5 μm),以磷酸盐缓冲液(pH=7.3~7.7)-乙腈(30+70,V1+V2)为流动相,进样量为10 μL,流速为0.6 mL/min,检测波长为195 nm,柱温为35 ℃。在此色谱条件下,葡萄糖胺盐酸盐在3.00~5.00 mg/mL浓度范围内呈良好线性关系,r = 0.99951,方法平均回收率为99.1%,相对标准偏差(RSD)为0.7%(n=9)。本法操作简便、快速、结果准确,应用于实际样品检测,结果满意,可作为葡萄糖胺盐酸盐含量测定的质控方法。

Abstract:
A method for determination of glucosamine hydrochloride in feed material by HPLC was developed. The separation was performed on ODS C18 column(150 mm×4.6 mm,5μm) with mobile phase of phosphate buffer solution(pH=7.3~7.7) - acetonitrile (30+70,V1+V2) and flow rate was 0.6 mL/min. The determination was done at 35 ℃ for the column temperature and at 195 nm for the detect wavelength. Under above experimental conditions, the calibration curve for glucosamine hydrochloride was linear in range of 3.00 ~ 5.00 mg/ml (r = 0.99951). The average recovery was 99.1%(n=9, RSD=0.7%). The results obtained by experimental verification showed the method is simple, rapid and reliable for the determination of glucosamine hydrochloride in feed material with satisfactory results.
