高效液相色谱法检测多种动物组织中噁喹酸和氟甲喹残留量的研究

摘要:
建立了猪、牛、羊的肌肉、脂肪、肝脏和肾脏四种组织中噁喹酸、氟甲喹残留量检测的高效液相色谱法。试样经磷酸盐缓冲溶液提取,C18固相萃取柱浓缩净化,用高效液相色谱分析,荧光检测器检测,外标法定量。动物组织中噁喹酸和氟甲喹的检测限为10 μg/kg,定量限为20 μg/kg。噁喹酸、氟甲喹在3~500 ng/mL范围内呈良好的线性关系。在20~300 μg/kg的浓度添加水平上,其平均回收率为63%~100%,批内、批间的相对标准偏差均小于15%。

Abstract:
An HPLC method for the determination of oxolinic acid and flumequine in muscle,fat,liver and kidney of swine,cattle and sheep has been developed.The sample residues were extracted by phosphate solution,purifid with C18 solid phase extraction column,seperated by HPLC with a fluorescence detector and quanlified by external standard calibration curves.The limit of detection was 10 μg/kg foroxolinic acid and flumequine,with quantification limit of 20 μg/kg.A good linearity of the calibration curves for oxolinic acid and flumequine was obtained within the range of 3~500 ng/mL.The average recoveries at level of 20~300 μg/kg were 63%~100%.The intra-and-interbatch variation coefficients were both less than 15%.
