兽药氟尼康中主要成分的高效液相色谱分析

摘 要:目的 建立HPLC法同时测定药物氟尼康中盐酸多西环素、氟苯尼考的含量。方法 采用Hypersil-ODS2 (250 mm×4.6 mm,5 μm)色谱柱,流动相为乙腈:甲醇:磷酸二氢铵(0.1 mol•L-1)=1:2:5,pH=3.50,检测波长为254 nm,流速为1 mL•min-1。结果 实现了氟尼康中盐酸多西环素、氟苯尼考的较好分离;盐酸多西环素在6.25 μg•mL-1~200 μg•mL-1范围内线性关系良好,r=0.9994,平均回收率为99.29%,RSD为0.51%;氟苯尼考在6.25 μg•mL-1~250 μg•mL-1范围内线性关系良好,r=0.9999,平均回收率为98.87% ,RSD为0.32%。结论 该方法快速简便,灵敏度高,重现性好,可用于药物氟尼康质量控制的测定方法

Abstract:
ABSTRACT: OBJECTIVE To establish a HPLC for the simultaneous determination of doxycycline hydrochloride and florfenicol in veterinary drug funicon. METHODS The liquid Chromatography was carried out by a Hypersil-ODS2(4.6 mm×250 mm,5 μm).The mobile phase was acetonitrile,methanol and 0.1mol•L-1 ammonium phosphate monobasic of pH 3.5(1:2:5) and the flow rate was 1mL•min-1. Funicon samples were measured with UV detector at 254 nm. RESULTS This experiment can get a better result of separation between doxycycline hydrochloride and florfenicol in funicon.The linear range of doxycycline hydrochloride was 6.25 μg•mL-1~200 μg•mL-1 and florfenicol was 6.25 μg•mL-1~250 μg•mL-1 (r=0.9999).The average recovery of doxycycline hydrochloride was 99.29%,relative standard was 0.51% (n=3). The average recovery of florfenicol was 98.87%,relative standard was 0.32% (n=3). CONCLUSION The method is simple,rapid and sensitive with good repeatability,can be used for content determination of funicon. KEY WORDS: HPLC; doxycycline hydrochloride; florfenicol; funicon
