狂犬病作为一种古老且致命的人畜共患病,至今仍对全球公共卫生安全构成严重威胁。疫苗接种是阻断病毒传播的核心策略,而mRNA疫苗凭借其快速研发潜力、高效免疫原性及灵活的 生产工艺,为狂犬病防控提供了新方向。聚焦狂犬病mRNA疫苗研发进展,系统阐述了mRNA疫苗的分子设计原理和递送体系优化策略,结合临床前研究与I期试验数据介绍了狂犬病mRNA疫苗的免疫原性特征及其在病毒中和与长效免疫记忆中的潜在优势,从稳定性、长期安全性和免疫原性的个体差异等方面探讨了狂犬病mRNA疫苗面临的挑战并提出建议,相信狂犬病mRNA疫苗将在实现2030年全球消灭狂犬病目标中发挥重要战略价值。 |

Abstract: |
Rabies, as an ancient and fatal zoonotic disease, still poses a serious threat to global public health safety.Vaccination is the core strategy to interrupt the transmission of the virus, and mRNA vaccines provide a new direction for the prevention and control of rabies due to their rapid development potential, highly efficient immunogenicity, and flexible production process.Focusing on the progress of rabies mRNA vaccine research and development, the molecular design principle and delivery system optimization strategy of mRNA vaccine were systematically explained;The immunogenicity characteristics of the rabies mRNA vaccine, including its potential advantages in virus-neutralizing activity and long-term immune memory, had been characterized through the integration of data from preclinical studies and Phase I clinical trials.The challenges faced by rabies mRNA vaccines were examined in terms of stability, long-term safety, and inter-individual variability in immunogenicity, with corresponding recommendations were also provided. It is believed that rabies mRNA vaccines will play a critical strategic role in achieving the 2030 global goal of rabies eradication. |
