研究旨在考察复合中草药对骨关节炎(OA)的治疗作用及其与硫酸软骨素的协同增效作用。试验选择30只健康SPF级雄性SD大鼠随机分为5组(假手术组、模型组和3个试验组),每组6只大鼠。适应性饲养1周后,假手术组大鼠关节腔注射0.9%生理盐水,其余各组大鼠注射碘乙酸钠(MIA)建立OA模型。造模第14 d,假手术组和模型组大鼠继续饲喂基础日粮,3个试验组分别在基础日粮中添加200 mg/(kg·d)硫酸软骨素、200 mg/(kg·d)复合中草药以及100 mg/(kg·d)复合中草药和100 mg/(kg·d)硫酸软骨素,给药4周。结果显示:造模后第28~42 d,试验3组大鼠机械刺激缩足阈值(PWT)、右下肢承重分布均显著升高(P<0.05);各试验组右膝关节股骨、胫骨缺损程度及关节间隙相比模型组均得到一定缓解。给药结束后,与模型组相比,试验2组和试验3组大体观察评分、Mankin评分均显著降低(P<0.05),大鼠关节软骨细胞数量有所增加,软骨基质染色增加;试验3组软骨中Ⅱ型胶原表达明显增加,滑膜中炎症因子白细胞介素-1β(IL-1β)表达明显减少。研究表明,复合中草药对OA大鼠的骨关节炎具有良好的治疗作用,联合硫酸软骨素具有增效作用,可为缓解动物OA的药物开发提供价值。 |

Effect of compound Chinese herbal medicine on osteoarthritis in rats |

Abstract: |
The purpose of this study was to investigate the therapeutic effect of compound Chinese herbal medicine on osteoarthritis (OA) and its synergistic effect with chondroitin sulfate. Thirty healthy male SD rats with SPF grade were randomly divided into five groups (sham operation group, model group and three experimental groups), with six rats in each group. After 1 week of adaptive feeding, the sham operation group was injected with 0.9% normal saline in joint cavity, and the other groups were injected with MIA to establish OA model. On the 14th day of manufacturing, the sham operation group and model group continued to feed the basic diet, and the three experimental groups were fed the basic diet supplemented with 200 mg/(kg·d) chondroitin sulfate, 200 mg/(kg·d) compound Chinese herbal medicine, 100 mg/(kg·d) compound Chinese herbal medicine and 100 mg/(kg·d) chondroitin sulfate, respectively. The drug was administered for 4 weeks. The results showed as follows: on the 28th to 42nd day after modeling, the mechanical stimulation foot contraction threshold (PWT) and the load-bearing distribution of the right lower extremity in test groups were significantly increased (P<0.05). Compared with the model group, the defect degree of femur and tibia of right knee and joint space in experimental groups were alleviated to some extent. After administration, compared with model group, the gross observation score and Mankin score of groups 2 and 3 were significantly decreased (P<0.05), and the number of articular chondrocytes and cartilage matrix staining of rats were increased. The content of type Ⅱ collagen in the cartilage of group 3 was significantly increased, and the content of inflammatory factor interleukin-1β (IL-1β) in synovium was significantly decreased. Studies have shown that compound Chinese herbal medicine has a good therapeutic effect on OA rats' osteoarthritis, combined with chondroitin sulfate has a synergistic effect, and can provide value for the development of drug to alleviate OA in animals. |
